Abstract: The cloning of less than microgram amounts of dsRNA of four plant viruses, cucumber mosaic virus (CMV), plum pox virus (PPV), raspberry leaf spot virus (RLSV), and strawberry mild yellow-edge virus (SMYEV), by using three methods of denaturation was accomplished. PPV and RLSV clones were up to 1,600 bp in length while the largest insert of SMYEV was 3,000 bp. Denaturation methods used were: boiling for 5 min; incubation in 90% dimethyl sulfoxide (DMSO) at 65 C for 30 min; and incubation at room temperature in 20 mM methylmercuric hydroxide (MeHg) for 10 min. The first strand and second strand synthesis were basically that of Gubler and Hoffman with the omission of mung bean nuclease digestion. After second strand synthesis, the DNA was size-fractionated on a Sepharose CL-4B column, C- tailed, annealed to G-tailed pBR322/pUC9, and Escherichia coli DH1/DH5a cells transformed. No differences in length of cDNA and size of inserts was observed when heat- and DMSO-denaturation were used. However, use of MeHg yielded greater incorporation of 32P-dATP into first strand when compared to the boiling method of denaturation.

Abstract: The pathogenicity of 12 isolates of Phytophthora fragariae was evaluated on 13 strawberry genotypes and then separated into seven distinct pathogenic races by their ability to infect and produce oospores in the roots of a set of differential strawberry hosts. The cultivars Aberdeen, Climax, Stelemaster, Surecrop, and Tennessee Beauty, MD683 selection, and the Fragaria chiloensis clone Del Norte are proposed as a new set of differential genotypes for identifying races of Phytophthora fragariae. A disease severity index based on the number of oospores in the root stele and the percentage of roots infected with P. fragariae is used to determine genotype susceptibility.

Abstract: The Japanese pear pathotype of Alternaria alternata, which causes black spot disease, produces multiple host-selective toxins (AK-toxins I and II) in culture; the strawberry pathotype, which causes black spot disease, also produces a series of analogous host-selective toxins (AF- toxins I, II, and III). Analysis by high-performance liquid chromatography of toxin production during spore germination showed that virulent isolate A85-10 of the Japanese pear pathotype produced about 0.02 pg of AK-toxin I per spore in the first 6 hr after germination and a trace amount of AK-toxin II after 24 hr. Most of the virulent isolates produced only AK-toxin I during the spore germination. Isolate YM-19 of the strawberry pathotype released about 0.08 pg of AF-toxin I per spore in the initial 6 hr of germination and a small but detectable amount of AF- toxin II, but no detectable AF-toxin III, within 18 hr of incubation. From the point of view of pathological importance of early host-parasite interactions, we propose that AK-toxin I and AF-toxin I, respectively are the major toxins in yield and biological activity which play a critical role as host recognition factors at primary infection sites in each pathosystem.

Abstract: Effects of ground cover, plant canopy density, and rain intensity on the splash dispersal of Colletotrichum acutatum were studied using a rain simulator. In one experiment, three ground covers (soil, straw, and plastic) and two rain intensities (15 and 30 mm/hr) were evaluated by collecting splash droplets with conidia in sheltered gravity samplers consisting of petri plates with a selective medium for C. acutatum. Ground covers were characterized by random roughness, the standard deviation of surface elevation. Infected fruits with sporulating lesions were clustered on the ground to serve as the inoculum source. Sampling plates were positioned 20, 40, 60, 80, 100, and 120 cm from the source and were exposed to rain for 1-min periods (every 5 min) for a total duration of 46 min. Ground cover had a major effect on splash dispersal, as measured by colonies growing in selective medium. The estimated total number of colonies over time and space (N) was inversely proportional to roughness; i.e., straw had the largest random roughness but the lowest N, and plastic the opposite. Differences in N among ground covers were due to differences in steepness of the dispersal gradients (i.e., straw had the steepest gradient and plastic the shallowest), not to the release rate of spores at the source. Total number of colonies increased with rain intensity, but the effect was due to the release rate, as measured by the intercept parameter of a gradient model. Rain intensity did not influence gradient steepness. The effect of plant canopy density on splash dispersal of C. acutatum was evaluated in a second experiment using a soil ground cover. Leaf area index (LAI) was inversely related to N. Cross-row dispersal of spores was reduced by 90% or more for two rows of plants with LAI greater than or equal to 2.7 compared to no plant rows. Results support the hypothesis that surface topography (including ground cover and plant canopy) is a major factor controlling splash dispersal due to its effect on splash droplet trajectories and loss of inoculum.

Abstract: This study was designed to determine if the cost of soil fumigation would be recovered in the first year of production when yield potential was being limited by the strawberry root rot complex. Yield, stand counts, leaf mineral nutrient levels, and nematode population levels, were evaluated for each of three treatments consisting of 67% methyl bromide plus 33% chloropicrin (MC33), 98% methyl bromide plus 2% chloropicrin (MC2) and a nonfumigated control. Yield was significantly greater in the fumigated plots than the control plots but did not differ significantly between the two fumigants. Stand counts and leaf mineral nutrient levels were not found to vary significantly among treatments. Compared with the control, fewer plant pararsitic nematodes were observed in plots treated with either MC33 or MC2. Nematode population differences between the MC33 and MC2 treatments were not evident. The cause of the black root rot complex was not determined from this study. Soil fumigation with either fumigant increased yields enough to justify this practice economically.

Abstract: Immature and mature strawberry fruit were inoculated with a conidial suspension (2.5 X 10(4) conidia/ml) of Colletotrichum acutatum and incubated under various wetness (free water) durations ranging from 0.5 to 51 hr at constant temperatures of 4, 6, 10, 15, 20, 25, 30, and 35 C. After drying, plants were moved to a greenhouse where incidence of fruit infection was recorded daily for 8 days. No infection occurred at 4 or 35 C on immature fruit or at 4 C on mature fruit. Generally, fruit disease incidence increased with increased wetness durations, but on mature fruit at 35 C, disease incidence decreased over time from a maximum of 39%. Optimum temperature for infection on both immature and mature fruit was between 25 and 30 C, with greater than 80% disease incidence after 13 hr of wetness. A regression model using the logit of disease incidence as the dependent variable accurately described infection level as a function of wetness duration (W) and temperature (T). Terms in the model were W, WT, WT2, and WT3, and all estimated parameters were significant. Coefficients of determination for combined data from three repetitions of the experiment were 0.71 and 0.83 for immature and mature fruit, respectively.

Abstract: Experiments were conducted to estimate the relative importance of additive and dominance genetic variances and non-allelic interactions in the inheritance of resistance to Colletotrichum spp. in strawberry (Fragaria X ananassa Duch.). Progeny of 40 parents crossed in a Comstock and Robinson Design II Mating scheme were inoculated with three isolates of C. fragariae and one isolate of C. acutatum. Disease development on each plant was rated visually. Variance components were estimated and converted to genetic variances. Estimates of dominance genetic variance were six to 10 times higher than those for additive genetic variance. Within-family variance not accounted for by additive and dominant genetic variance equaled 35% and 38% of the total genetic variance in females and males, respectively, indicating probable epistatic effects. The frequency distribution of disease severity ratings was bimodal in both experiments, suggesting major gene action. Narrow-sense heritability estimates were 0.37 and 0.26, and broad-sense heritability estimates were 0.87 and 0.85 for females and males, respectively. Narrow-sense heritability estimates are probably sufficient to produce gains from recurrent selection. Gains from selection of clonal value should be possible because of the high broad sense heritability estimates. It appears feasible to establish a broad genetic-based population resistant to Colletotrichum spp. from which selections could be evaluated per se and/or recombined to produce improved populations.

Abstract: Pratylenchus penetrans was obtained from Premier strawberry in Norfolk County and the Niagara Peninsula and from celery in the latter area. Host affected the dimensions of P. penetrans to a greater extent than geographical area in Ontario. Adults of P. penetrans from southern Ontario tended to be smaller than those reported elsewhere. The presence of three lip annules was consistent in the seven populations studied, although in some specimens one of the annules did not entirely encompass the head. Crenations around the tail tip of females of P. penetrans was common in the populations studied. Not all of the morphological characters were proportional in size to length of the females of P. penetrans.

Abstract: Strawberry crinkle virus (SCV), a cytoplasmic rhabdovirus, has proven difficult to purify from strawberry leaves. By testing alternative hosts and different procedures, we have succeeded in purifying SCV from leaves of Physalis floridana using a celite filtration method. Two classes of particles differing in sedimentation rate could be distinguished by sucrose density gradient centrifugation. Electron microscopic examination of these purified preparations revealed the presence of bacilliform particles and an apparent size difference between the two classes. Polyacrylamide gel electrophoresis of sodium dodecyl sulfate-disrupted virions showed that SCV contains three major structural proteins with molecular weights estimated to be 78, 47, and 25 kDa. The 78-kDa protein bound lectins, identifying it as the viral glycoprotein. The SCV protein profile resembles that of lettuce necrotic yellows virus (LNYV) but differs appreciably from sonchus yellow net virus (SYNV). SCV has no detectable serological relatedness to LNYV or SYNV. These results suggest that SCV is distinct from other previously characterized rhabdoviruses.

Abstract: A rain simulator was used to investigate the influence of rain intensity (15 and 30 mm/hr), rain duration (15-60 min), and ground cover on splash dispersal of Colletotrichum acutatum from infected strawberry fruit. Potted strawberry plants were held in two concentric circles (30- and 60-cm radii) by a wood frame which was levelly exposed to a uniform zone of generated rain. Infected fruits with sporulating lesions were placed in the center of the circles. Studies were conducted with three ground covers: soil, soil covered with fresh straw (6-8 cm deep), and plastic. With plastic cover, 100% disease incidence was obtained at both distances and all times. With soil and straw, disease incidence was generally less in the outer compared with the inner circle, but increased in both circles over time only with soil. With 60 min of rain at 30 cm from the source, mean disease incidence was greater than or equal to 90%. Rain intensity, however, did not have a consistent effect on dispersal. Influence of number (source strength) and distribution of infected fruit on resultant dispersal was evaluated, with one, five, or nine infected fruits clustered in the center of the circles. or five or nine fruits uniformly scattered over the frame. Neither source fruit number nor its spatial distribution, however, was found to significantly affect disease incidence. A simple wind tunnel also was incorporated into the study to evaluate the effect of horizontal air flow. With a flow of approximately 2.3 m/sec, differences in disease incidence between up- and downwind locations were significant at 60 cm from the source but not at 30 cm. At 60 cm, disease incidence downwind (approximately 60%) was triple the incidence upwind (approximately 18%).

Abstract: Several strains of Agrobacterium tumefaciens and A. rhizogenes were shown to form tumors on runners of the diploid strawberry species Fragaria vesca L. Tumors, weighing from 0.1 to 8.3 mg, appeared from 2 to 4.5 weeks after infection. The majority of tumors tested for opine synthesis by high-voltage paper electrophoresis analysis showed positive results. These results demonstrate that diploid strawberry plants are susceptible to infection with Agrobacterium and that there are differences in the relative virulence of Agrobacterium strains.

Abstract: Strawberry (Fragaria X ananassa Duch.) seedlings were planted in a greenhouse at 3- to 4-week intervals and simultaneously inoculated at ages 80, 101, 129 or 157 days with either of two naturally occurring virus sources each of which contained a mottle, mild yellow edge, and crinkle virus complex. Inoculation by aphids with either virus source reduced vigor, petiole length, leaflet width, stolons per plant, and vegetative dry weight of plants in the greenhouse. The tendency of virus inoculation to reduce vigor and petiole length was inversely proportional to increasing seedling age. In the field, inoculated seedlings were also less vigorous than control seedlings. Virus source effects and seedling age interactions with virus source were not significant. Selection for virus tolerance, based on greenhouse vigor, petiole length or leaflet width measurements, increased the frequency of seedlings subsequently classified as virus-tolerant in the field in both 80- and 101-day- old seedlings. Selection based on greenhouse vigor or petiole length increased the frequency in 129-day-old seedlings. No greenhouse selection method evaluated was effective in 157-day-old seedlings.

Abstract: Characteristics of splash dispersal of Phytophthora cactorum from infected strawberry fruit were studied with an integrated drop- generating and photographic system that was developed previously. Drops of 1- to 4-mm diameter were released from heights of 10, 20, and 40 cm above fruit bearing sporangia that had been labeled with a fluorescent tracer. Splash droplets produced by the impact of the incident drops were collected on sheets of water-sensitive paper. Drops with diameters of 1 and 2 mm failed to produce any splash droplets when released from heights of 20 and 10 cm, respectively. Multivariate regression analysis was used to examine the relationship between a set of intercorrelated responses and attributes of the incident drop. Responses evaluated were 1) the total number of droplets collected, 2) the average droplet diameter, 3) the total droplet volume, 4) the mean distance of droplet travel, 5) the number of droplets bearing at least one sporangium, and 6) the total number of sporangia among droplets with a least one sporangium. Incident drop attributes were drop mass or weight (W) and the velocity (V), momentum (M), and kinetic energy (E) of the drop at impact with the fruit surface. The best multivariate regression model included W, M, and V as predictors of reponses 1, 2, and 4. M alone was the best predictor of response 3; in constrast, V was significant alone for responses 5 and 6. Except for droplets produced by the impact of 1-mm-diameter drops, the average diameter of droplets carrying sporangia was always significantly greater than the diameter of droplets not carrying sporangia (P less than or equal to 0.05). The distribution of the number of sporangia per droplet was generally well described by the logarithmic distribution with zeros for each combination of incident drop diameter and height of release. The gamma parameter, representing the proportion of droplets with sporangia, declined in relation to momentum at impact. The theta parameter, a measure of the mean number of sporangia per droplet (of droplets bearing sporangia), declined with increasing mass of the incident drop. There was no relation between droplet size and the number of sporangia per droplet. For all splash droplets and those with sporangia, the distribution over distance was accurately described by the Weibull distribution whose scale parameter (representing the distance within which 63% of the droplets landed) varied linearly and positively with incident drop velocity. The Weibull shape parameter (representing curve skewness) was best described as a function of both drop velocity and kinetic energy or momentum. Droplets with sporangia did not travel as far, on the average, as droplets without sporangia.
