Abstract: The objective of this study was to determine the influence of the number of visits by the honey bee (Apis mellifera L.) on the pollination rate of strawberry (Fragaria x ananassa Duch.) flowers. Pollination rate is defined as the number of fertilized achenes divided by the total number of achenes per berry. Veestar' cultivar plants were caged before flowering began and cages were opened to observe bee visits. The cumulative effect of the number and length of visits to these flowers significantly increased the pollination rate. Four visits with a total of approximately 40 s of foraging were required for adequate pollination. In the absence of pollinators, primary flowers had lower pollination rates than secondary and tertiary flowers. Pollen transported by bees is thus particularly important for optimal development of these first berries harvested for market.

Abstract: Nitrogen and carbohydrate content of roots and the onset of crown cold hardiness were compared in 'Tristar' day-neutral (DN) strawberries (Fragaria X ananassa Duch.) that were given various fruit removal treatments. Nontreated 'Hecker' DN and 'Redcoat' Junebearer were also used to determine genotypic variation. The removal of fruit after 15 or 30 Sept. promoted the accumulation of starch and increased cold tolerance of crowns as compared to fruiting plants. Nitrogen was increased only when fruit was removed after 15 Sept. DN cultivars were less hardy than Junebearing cultivars, but 'Tristar' was almost as hardy as 'Redcoat'. When compared to 'Redcoat', DN cultivars had a less abrupt temperature- kill profile, perhaps because they were multicrowned.

Abstract: Hermaphroditism is the normal mode of sex expression in diploid species of Fragaria (Rosaceae, 2n = 14, x = 7) with one known exception, gynodioecious F. vesca L. ssp. bracteata. The polyploid species of Fragaria are all trioecious. An extensive study involving appropriate hybridization, testcrossing, selfing, and backcrossing revealed that the repression of sporangia and the inhibition of sporogenesis are controlled by a single gene (or a gene complex) with at least three alleles in the sporophytes of trioecious species (2n = 56, x = 7). A male suppressor (allele F) reduces microsporangia and represses completely, but it allows normal development of the stigma, style, and ovary. A female suppressor (allele M), in the absence of modifier genes, inhibits and drastically reduces the number of carpels and size of receptacles. The allele H, conferring perfect flowers at an early stage of flower organogenesis, acts as an inducer of microsporogenesis in females, but leaves both micro-and megasporangial development intact. At the sex locus, the F allele (femaleness) is dominant to H and M and the H allele (hermaphroditism) is dominant to M (maleness). Females are exclusively heterogametic (F/H or F/M), hermaphrodites may be homo- or heterogametic (H/H or H/M), and males are homogametic (M/M). The sex gene is expressed precisely in the genetic background of X diploid hybrids of Fragaria and their derivatives and in crosses with closely related hermaphroditic diploid Pontentilla glandulosa L. Gene dosage phenomena are absent. First generation progeny of colchi-decaploids (F/F,-/-) are exclusively female, but all generations thereafter segregate in a normal diploidized manner (1:1). Application of phytohormones alters sex expression to a limited extent.

Abstract: Concentrations of foliar phenolics varied significantly between clones of Fragaria chiloensis (L.) Duchesne and Fragaria X ananassa 'Totem' and among leaves of different ages within an individual plant. In contrast, qualitative analysis of phenolic compounds, determined with high-pressure liquid chromatography, revealed a close similarity between clones of F. chiloensis and F. X ananassa 'Totem.' Development of the twospotted spider mite, Tetranychus urticae Koch, was negatively correlated to concentrations of foliar phenolics; mite development was delayed on leaves with high concentrations of phenolics. Catechol-based phenolics were more strongly correlated with mite development than were total foliar phenolics.

Abstract: Mixing strawberries ground under liquid nitrogen with polyvinyl-pyrrolidone alone or combined with Amberlite XAD-4 in pH 4.5 buffer yielded stable polyphenoloxidase (PPO) extracts. Temperature stability of the enzyme varied with substrate and followed first order kinetics of inactivation. pH optima were 5.5 with catchol and 4.5 with 4-methylcatechol. Maximum activity was with D-catechin, followed by 4- methylcatechol and pyrogallol with no cresolase activity. Inhibition occurred with diethyldithiocarbamate, potassium metabisulfite, KCN and dithiothreitol. Benzenesulfinic acid, cysteine and ascorbic acid blocked the browning reaction without enzyme inhibition. Extraction with Dowex AG 2-X8 resulted in browning. No loss in activity after 95 days occurred with quick-frozen fruit stored under nitrogen at -40 degrees C.

Abstract: Stable and highly active polyphenoloxidase (PPO) extracts were obtained using polyvinylpyrrolidone, Amberlite XAD-4, Triton X-100, and protease inhibitor in pH 5.25 buffer. Citrate and phosphate prevented binding of PPO to pectin. Phenyl sepharose chromatography resolved PPO into two isozymes (PPO-F1 and PPO-F2) both separated from pectin. PPO-F1 had a molecular weight of 111,000, did not penetrate the running gel during electrophoresis, and was bound by concanavalin suggesting that it contained carbohydrate. PPO-F2 resolved into two peaks during DEAE-chromatography, had a molecular weight of 34,500, and resolved into two bands during electrophoresis.

Abstract: Xylem sap from apple (Malus domestica Borkh), peach (Prunus persica Batsch), and pear (Pyrus communis L.) twigs was collected by means of pressure extrusion. This sap contained a number of acidic peroxidases and other proteins. Two other sources of xylem sap used in this study were stem exudates and guttation fluid. Similar peroxidases were also found in stem exudates and guttation fluids of strawberry (Fragaria X ananassa Duch.), tomato (Lycopersicum esculentum L.), and cucumber (Cucumis sativus L.). Isoelectric focusing activity gels showed that two peroxidases (isoelectric point L 9 and pl 4.6) were present in initial stem exudates collected in the first 30 minutes after excision. Subsequent samples of stem exudate collected contained only the pl 4.6 isozyme. The pl 4.6 peroxidase isozyme was also found in root tissue and guttation fluid. These observations suggest that roots produce and secrete the pl 4.6 peroxidase into xylem sap. Cucumber seedlings were treated with 100 microliters per liter ethylene for 16 hours and the exudate from decapitated hypocotyl stumps was collected over a 3 hour period. Ethylene increased the peroxidase activity of stem exudates and inhibited the amount of exudate released. These observations suggest that xylem sap peroxidase may play a role in plugging damaged vascular tissue.
