    • Journal of Microbiological Methods
    • Author(s): Julio C. Ayala, Jorge A. Benitez, Anisia J. Silva

      Abstract

We describe a proteomic approach to identify transcription factors binding to a target promoter. The method's usefulness was tested by identifying proteins binding to the Vibrio cholerae rpoS promoter in response to cell density. Proteins identified in this screen included the nucleoid-associated protein Fis and the quorum sensing regulator HapR.

  11. Chloroform-free permeabilization for improved detection of β-galactosidase activity in Vibrio cholerae

    • Journal of Microbiological Methods
    • Publication date: Available online 21 March 2017

Author(s): Charlotte Toulouse, Claudia C. Häse, Julia Steuber

LacZ (β-galactosidase) is used to monitor the transcription of genes in reporter strains carrying the lacZ gene under the control of a promotor of interest. This protocol for LacZ activity determinations in Vibrio cholerae following detergent lysis results in 2.5-fold increase of LacZ activities compared to lysis with chloroform.

We evaluated the utility of an immunochromatographic assay (NH IC TDH) in identifying thermostable direct hemolysin (TDH)-producing Vibrio parahaemolyticus colonies on selective agar plates. The sensitivity of the NH IC TDH assay was 100% (189 samples) and its specificity was 100% (41 samples) compared with the presence of tdh.
