Detection of Chemical Substances in Bovine Liver, Rumen Contents, and Serum as Determined by GC/MS

Objective

To identify all of these compounds using mass spectrometry and also their relative retentions times by GC/MS analysis. With a complete list of expected compounds, it will be easier to quickly and definitively identify any foreign or abnormal xenobiotics compounds and metabolites that may be detrimental to animal health or food safety.

NON-TECHNICAL SUMMARY: Bovine liver, rumen contents, and serum are commonly submitted to the PADLS Toxicology Laboratory to be analyzed for the presence of potentially harmful substances that may compromise both animal health and food safety, such as pesticides, environmental contaminants, and pharmaceutical agents. Because of the nature of biological specimens, many complex yet clinically normal compounds are expected to be detected, but a formal survey of these substances has not previously been reported. Determination of the normal organic profile of these various matrices would facilitate better diagnostic interpretation of the analyses of these sample types for the detection of substances potentially dangerous to animal and human health. <P>

APPROACH: A minimum of 20 samples each of bovine liver, serum, and rumen contents will be analyzed by our GC/MS organic chemical screen using diphenylamine as an internal standard. The molecular weight, retention time, and the name (if available) of each novel compound will be added to the GC/MS library search for future reference. A comprehensive list of the compounds will be prepared and the relative ratio of each compound to diphenylamine will be measured and tabulated.
