The purpose of this project is to develop a robust DNA preparation method that can be integrated into the Lab existing multiplex PCR/microarray-based assay for simultaneous detection of nine bacterial and parasitic pathogens in foods in a single test. The proposed method is based on the new multiple displacement amplification (MDA) technology that employs 29 DNA polymerase and exonuclease-resistant primers for faithful replication of the original DNA population. Procedures for DNA isolation and enrichment by MDA will be developed for various foods. It will be explored whether the MDA method can be directly used with microarray without PCR.
Expected Impact of Project Outcomes on Food Safety in Ontario: The deliverable of the project will be a robust DNA extraction and enrichment method for food samples, including poultry, red meat, milk and vegetables, which can be used as a front end for PCR/microarray-based assays. The availability of such a DNA preparation method (a front end) will allow diagnostic labs to apply the powerful microarrays in food testing to provide faster and more cost-effective results to food industries and government agencies and provide an avenue to enhance the safety of Ontario's food supplies.

This will also allow scientists to conduct risk assessment studies in a systematic and cost-effective manner in support of monitoring and surveillance activities and enforcement of regulatory programs. This will help to ensure that the Ontario agri-food industries remain competitive in providing safe foods to today's food safety conscious consumers.
